Standard protocol for monoclonal antibody development service includes the 5 phases:
Phase 1: Antigen preparation:
- Protein antigens: customers provide high purified protein antigens (5 to 8 mg for immunization; 2 to 4 mg for ELISA screening; 1 to 2 mg for characterization by western blotting).
- Peptide antigens: customers provide peptide-carrier protein conjugates. Customers provide at least 4 mg peptide conjugated to KLH for immunization and at least 2 mg peptide conjugated to different carrier protein, e.g. OVA or BSA for ELISA screening and western blotting characterization. Instead, customers can provide peptide sequence; we synthesize peptide and prepare the peptide-carrier conjugates (get quote).
Phase 2: Immunization and ELISA titration ($2500)
Five BALB/c mice will be immunized by a standard protocol (can be adjusted base on costumer’s experimental information and request). We will optimize ELISA assay for antibody titration and hybridoma screening. We will send antibody titration data to customer for evaluation.
Phase 3: Cell fusion, screening, subcloning and characterization by western blotting ($2500)
Two best-immunized mice will be selected for cell fusion and preparation of hybridoma. Hydridoma cells will be selected with HAT selection medium. Supernatants from hybridoma cells will be screened by ELISA. At least 5 best parental hybridoma lines will be subcloned to ensure monoclonal line. 5 stable lines will be further characterized by western blotting. Supernatants (5 ml from each of the selected single clone hybridoma) will be sent to customer for evaluation.
Phase 4: Expansion, freeze parental stocks and delivery ($1000)
Three hybridoma cell lines from different parental hybridomas will be further expanded and frozen-down. Six frozen vials (2 from each of 3 parental lines) will be delivered to customers.
Phase 5: Monoclonal antibody production and purification ($400)
Monoclonal antibody will be purified from ascites or hybridoma supernatants by Protein A/G affinity chromatography. 9 mg (3 mg from each of the 3 customer-selected clones) will be delivered to customer.
